Acupuncture activates GABA neurons in the thalamic reticular nucleus to regulate sleep-wake cycles in PCPA-induced insomnia in mice

Acupuncture reduced sleep latency and increased sleep duration in PCPA-induced insomnia mice (P < 0.01). It increased Ca2+ signals in GABA neurons within the TRN (paired t-test, P < 0.01) and shifted EEG power from elevated Beta/Gamma frequencies to increased Delta power (P < 0.01). Chemogenetic inhibition of TRN GABA neurons blocked these effects.

Frequently Asked Questions

What is the main finding of this study?โ–ผ

Acupuncture activates GABA neurons in the thalamic reticular nucleus, which helps regulate sleep-wake cycles and improves insomnia in a mouse model.

How was insomnia induced in the mice?โ–ผ

Insomnia was induced using p-chlorophenylalanine (PCPA), a compound that depletes serotonin and disrupts sleep.

What measurements were used to assess sleep improvement?โ–ผ

Wheel-running activity, sodium pentobarbital-induced sleep tests, fiber photometry for Ca2+ signals, and cortical EEG recordings were used.

What happened when GABA neurons in the TRN were inhibited?โ–ผ

Chemogenetic inhibition of TRN GABA neurons blocked the beneficial effects of acupuncture on EEG power, suggesting these neurons are essential for the treatment effect.
